Isogenic iPS Cell Lines
Alstem provides a suite of isogenic iPSC-derived cell lines specifically engineered for studying the role of APOE in neurodegeneration. Starting from an APOE ε4 iPSC line, generated from human bone marrow CD34+ mononuclear cells reprogrammed with transient Yamanaka factor expression, we have created a panel of isogenic APOE variants (ε2, ε3, and knockout) using CRISPR-mediated gene editing. In addition, an inducible NGN2 version has been established, with NGN2 integrated into the AAVS1 safe harbor locus, enabling robust and reproducible induction of excitatory neurons. These isogenic models provide a powerful platform to dissect APOE-associated mechanisms and accelerate translational research in Alzheimer's disease and related neurodegenerative disorders.
